S100A13基因的高表达对HepG2细胞生物学性状影响的研究

摘    要:目的 观察S100A13基因的高表达对HepG2细胞生物学性状的影响.方法 采用以绿色荧光蛋白(GFP)为报告基因的重组表达质粒PcDNA3.1/NT-GFP-S100A13,经脂质体介导转染肝癌细胞系(HepG2);荧光显微镜观察和RT-PCR鉴定表达S100A13的细胞克隆并检测转染前后细胞FGF1表达量的变化;MTT观察阴性对照组、转染空载体组和转染重组基因组的细胞生长曲线.各组分别加入化疗药物(5-Fu),24h后收集细胞,流式细胞术检测细胞凋亡.结果 转染后的重组细胞系S100A13表达明显增高,转染PcDNA3.1/NT-GFP-S100A13组FGF1的表达量明显高于阴性对照组及空载体组.但各组细胞的生长曲线以及对化疗药物(5-Fu)的敏感性差异无显著性.结论 S100A13及其介导释放的FGF1在肿瘤的增殖及对抗癌药物的敏感性方面均没有明显的作用.

Abstract:[Objective] To research the biological characterization of HEPG2 cell line transfected with S100A13 gene.[Methods] The recombinant PcDNA3.1/NT-GFP-S100A13 was transfected into HEPG2 cells by lipofectamine,the mRNA expression level of S100A13 and FGF1 were identified by RT-PCR,respectively.The cell proliferation status was examined by MTT technical.Then cells were treated with 5-Fu.The cell apoptosis rates were detected by flow cytometry.[Results] S100A13 was stably expressed in transfected cell line and the...
