华佗再造丸对大鼠局灶性脑缺血损伤的保护作用

摘    要:目的 探讨华佗再造丸(HTZZ)对大鼠局灶性脑缺血再灌注损伤后缺血半暗带的神经元及胶质细胞的保护作用。方法 采用大鼠局灶性大脑中动脉阻断(MCAO)模型,于缺血再灌注(I/R)后第22、70小时分别进行神经功能评分(NDS);TTC染色测定脑梗死体积;免疫组织化学的方法测定烯醇化酶(NSE)和胶质纤维酸性蛋白(GFAP)的动态变化。结果 HTZZ治疗组与对照组相比,在大鼠I/R后22、70 h NDS均明显增高(P<0.05);脑梗死体积明显缩小(P<0.05);NSE的表达明显增强(P<0.05);GFAP的表达均较对照组明显减少(P<0.05);在I/R后70 h GFAP的表达较22 h时略有增加(P>0.05)。结论 HTZZ对大鼠局灶性脑缺血再灌注损伤后缺血半暗带的神经元及胶质细胞有显著的保护作用,是一种有效的神经保护剂。

Abstract:OBJECTIVE To investigate the protective effects of Huatuo Zaizao pills(HTZZ) to neuron and glial cells in the "penumbra" area on focal cerebral ischemia-reperfusion(FCI- R) injury in rats. METHODS The FCI - R model with thread embolism of middle cerebral artery occlusion (MCAO) in rats was made.The neurological deficit score(NDS) was assessed.TTC staining were used to evaluate the infarct volume of injured rats. Adopting immunohistochemistry method to detect the changes of neuron - specific enolase (NSE) and glial fibrillary acidic protein (GFAP) expression at the time of 22 h,70 h after MCAO. RESULTS Compared with those in the control group, the NDS were all significantly increased( P < 0.05) ;the infarct volume were significantly decreased(P < 0.05) ;the level of NSE expression in the "penumbra" area were all significantly increased( P < 0.05) and the level of GFAP expression in the "penumbra" area were significantly decreased( P < 0.05 ) in the HTZZ - treated group at 22 h, 70 h after FCI - R ( P < 0.05 ) . The level of GFAP expression at 70 h after FCI - R were slightly increased ( P > 0.05)than that at 22 h after FCI - R in the HTZZ - treated group. CONCLUSION The result shows that HTZZ has significant protective effects to neuron and glial cells in the "penumbra" area in FCI- R rats.
