摘    要:目的探讨生存素基因Survivin对缺血/再灌注神经细胞凋亡的影响。方法构建真核表达质粒pEGFP-Suvivin,脂质体介导转染细胞,筛选获得稳定表达Suvivin基因的转染细胞,于转染24h制作体外缺血/再灌注模型,于造模后6h对比模型组、空载体组、实验组细胞在Suvivin蛋白、细胞凋亡等方面的变化。观察Suvivin基因对缺血/再灌注神经细胞的抗凋亡作用。结果实验组细胞凋亡率明显低于模型组。结论Suvivin基因转染能有效地保护缺血/再灌注细胞免受凋亡,为其临床应用提供了理论和实验依据。

Abstract:Objective To research the effect of Survivin gene survivin to ischemia-reperfusion of nerve cells apoptosis. Methods To construct the eukarya expression recombinant pEGFP-survivin,, the bangosome mediate transfection cell, to select suvivin gene of transfection cell, at 24 h after transfection to make the model of ischemia-repeffusion, at 6 h after model to contrast the changes of Suvivin proteinum and apoptosis on model group, idler group and test group. To observe the anti-apoptosis effect of suvivin gene to ischemia-reperfusion nerve ceils. Results The apoptosis rate of test group was obviously lower than the model groups. Conclusion Survivin gene transfection can efficiently effect ischemia-reperfusion cells to against apoptosis, can provide theory and experiment base to clinical application.
