牛膝中齐墩果酸薄层扫描测定方法的研究

 采用薄层扫描测定牛膝中皂甙元齐墩果酸。对牛膝中皂甙的水解条件、齐墩果酸薄层分离及显色条件进行了研究，确定了用１ｍｏｌ／Ｌ硫酸沸水浴水解７ｈ，在自制含数甲基纤维素钠硅胶Ｇ薄层板，以氯仿－乙醚－甲醇（３０：１０：１）为展开剂分离，喷茴香醛－硫酸显色剂１００℃烘烤７ｍｉｎ显色等条件，测定了牛膝及其炮制品中齐墩果酸的含量。本方法的平均回收率为９８．６％，相对标准偏差为２．７％。

Determination of oleanolic acid in roots of Achyranthes bidentata byTLC-scan

A TLC-scan method was devdoped for determination of oleanolic acid in roots ofAchyranthes bidentata Bl..Influential factors of the saponin hydrolysis and visualization were ex-amined.The key experimental conditions were as follows:saponin hydrolysis at 100℃ with 1mol/L sulfuric acid for 7 h,TLC separation on a home-made silica gel G plate with chloroform-ethyl-ether-methanol（30:10:1）as a developer,visualization at 100℃ for about 7 min after spray-ing anisaldehyde-sulfuric acid reagent.Contents of oleanolic acid in the roots and the processedroots of Achyranthese bidentata were determined.The average recovery and RSD of the methodwere 98.3%and 2.7%respectively.
