正交试验法优选三七皂苷成分超声提取工艺

摘    要:目的:比较不同提取方法对三七皂苷成分含量测定的影响,优选三七皂苷成分提取工艺。方法:用HPLC同时测定三七皂苷R1、人参皂苷Rg,1、人参皂苷Rb。和人参皂苷Rd4种皂苷成分,并采用正交试验法,考察了甲醇浓度、超声时间、溶剂体积3个因素对三七4种皂苷成分提取率的影响,优选出合理的提取工艺。结果:采用超声提取法对三七中4种皂苷成分的提取效果同回流法基本~致。在超声提取工艺中,正交试验结果表明影响各种皂苷提取率的因素大小分别为超声时间(B)〉甲醇浓度(A)〉甲醇用量(C),其中甲醇用量对上述皂苷成分提取率影响可忽略不计。综合比较各种皂苷成分提取效率、操作周期和降低能耗等方面,超声提取的最佳工艺为A2B2C2,即称取0.30g三七药材,加80%浓度的甲醇25mL,放置过夜后,超声提取1h,超滤后用HPLC法同时检测分析4种单体皂苷成分含量。结论:所优选的工艺操作简便,重现性和稳定性较好,可作为大批量检测三七样品中4种皂苷成分含量的快速检测方法。

Abstract:Objective: To optimize the extraction conditions of Panax notoginseng saponins. Methods: We compared the influence of reflux and ultrasonic extraction on saponins test , and designed orthogonal experimental to investigate 3 factors which affect ultrasonic wave extraction. Notoginseng saponins were tested by HPLC at the same time. Results:There is no significant difference between reflux and ultrasonic wave extraction, but significantly impact on 3 experimental factors of ultrasonic wave extraction. The factors influence saponins extraction, first is ultrasonic time, second is methanol concentration and last is volume of solvent. The optimum condition is A2B2C2 ,which means the 0. 30 g sample with 25 mL 80% methanol processing 1 h by ultrasonic wave extraction. Conclusion: The results show that the technique is reliable and available for Panax notoginseng saponins fast test.
