蛇血清分泌型磷脂酶A_2抑制剂防治人炎症的生物信息学分析

摘    要:目的比较人和蛇毒分泌型磷脂酶A2(sPLA2)结构相似性,分析蛇血清磷脂酶A2抑制剂(PLI)对sPLA2特异性抑制的结构与功能关系,评价蛇血清PLI对人炎症的抑制作用。方法用Clustal W2对人和五步蛇sPLA2氨基酸序列进行多重序列比对。用Swiss-model对各种sPLA2的空间结构进行同源建模预测,并用Chimera软件对人和蛇毒sPLA2空间结构进行三维比对。结果人和五步蛇sPLA2一级序列同源性有40%左右,空间结构几乎相似。各种PLI具有相同的空间结构特征和保守的sPLA2结合区。结论理论上推断蛇血PLI可有效抑制人sPLA2活性,减少sPLA2介导的炎症发生。
