cNOS选择性抑制剂对兔桡骨缺损愈合影响的实验研究

摘    要:目的研究cNOS选择性抑制剂L-精氨酸甲酯对兔桡骨骨缺损愈合的影响。方法制备同种异体松质骨和cNOS选择性抑制剂L-精氨酸甲酯复合。制备骨缺损动物模型。左侧前肢植入同种异体松质骨,右侧前肢植入cNOS选择性抑制剂和同种异体松质骨复合物。术后2、4、8、12周,行肉眼观察、X线拍片、骨密度检查和组织学切片检查。术后12周,行三点弯曲试验。并与正常桡骨比较。结果单纯同种异体松质骨在骨缺损愈合过程中成骨量,成骨速度,时间均优于cNOS选择性抑制剂和同种异体松质骨复合物。结论cNOS选择性抑制剂L-精氨酸甲酯(L-NAME)可抑制骨缺损愈合。

Abstract:[Objective] To investigate the effect of selecctive inhibitor of cNOS(L-NAME ) on repairing radius bone defects in rabbit.[Methods] Allograft cancellous bone and compound with selective inhibitor of cNOS L-NAME were prepared.Bone defect was created on bilateral radius to make the animal models.The simple compound of cancellous allograft bone was implanted on left limb and the compound of cancellous allograft bone and selective inhibitor of cNOS L-NAME were implanted on right limb.The effect was observed by gross,histooathological X-ray examinations and the bone mineral density(BMD) at 2,4,8,12 week after operation.Mechanical strength was measured by three points curving test at 12 weeks after operation and compared with normal radius.[Results] The simple cancellous allograft bone was superior in terms of the amount and speed of new bone formed on region of bone defect to the compound of cancellous allograft bone and selective inhibitor of cNOS L-NAME.[Conclusion] Inhibitor of cNOS can inhibit repairing bone defect.
