不明原因早期复发性自然流产患者蜕膜组织中杀伤细胞免疫球蛋白样受体2DL1的表达

摘    要:目的研究不明原因早期复发性自然流产(RSA)患者早孕蜕膜组织中杀伤细胞免疫球蛋白样受体(KIR)2DL1 mRNA和蛋白的表达变化,探讨RSA发病的免疫机制。方法选择30例不明原因早期RSA者作为研究组,30例同期正常早孕要求行人工流产术者为对照组,采用SYBR绿色荧光染料I(SYBR Green I)实时荧光定量PCR法及Western blotting技术检测两组蜕膜组织中KIR2DL1 mRNA和蛋白的表达差异。结果① 研究组蜕膜中KIR2DL1 mRNA阳性表达率为90.00%(27/30),对照组蜕膜中KIR2DL1 mRNA阳性表达率为96.67%(29/30),两者无统计学差异(P>0.05)。研究组KIR2DL1 mRNA表达量用RQ值表示为699.37±259.78,对照组KIR2DL1 mRNA的表达量为1?002.96±476.976,差异亦无统计学意义(P>0.05);② 研究组蜕膜中KIR2DL1蛋白阳性表达率为86.67%(26/30),对照组蜕膜中KIR2DL1蛋白阳性表达率为93.33%(28/30),两者差异无统计学意义(P>0.05)。但研究组蜕膜中KIR2DL1蛋白的表达量0.28±0.14显著低于对照组0.71±0.14(P<0.05)。结论蜕膜组织中KIR2DL1表达量降低,传递给NK细胞的抑制性信号减少,可能影响母胎免疫耐受过程,导致RSA的发生。

Abstract:To investigate mRNA and protein expressions of a killer cell immunoglobulin like receptor(KIR) 2DL1 in deciduas of unexplained first trimester spontaneous abortion. MethodsExpressions of KIR2DL1 mRNA and protein in deciduas of unexplained first trimester spontaneous abortion and normal first trimester pregnant women were determined by SYBR Green I real time quantitative retroverse polymerase chain reaction (RT PCR) and Western blotting methods. Results① Expression of the KIR2DL1 mRNA in deciduas of the abortion group was less frequently than that of the control group (90.00% vs 96.67%,  P>0.05); Expressive quantity of the KIR2DL1 mRNA was lower in deciduas of the abortion group compared with that of the control group(P>0.05). ② Expression of the KIR2DL1 protein in deciduas of the abortion group was less frequent than that of the control group (86.67% vs 93.33%, P>0.05). Expressive quantity of the KIR2DL1 protein in deciduas of the abortion group was significantly lower than that of the control group(P<0.05). ConclusionDepressed expression of KIR2DL1 may correlate with occurrence of unexplained first trimester spontaneous abortion.
