半夏蛋白对小鼠的抗生育作用及抗早孕的机理探讨

摘    要:本文观察半夏蛋白对小鼠的抗生育作用并初步探讨了该药抗早孕的机理。实验表明半夏蛋白30mg/kg对小鼠具有明显的抗早孕作用,抗早孕率可高达100%。给药后24h就可见到血浆孕酮水平下降,在光镜下子宫内膜变薄,大部分区域已见不到蜕膜反应,胚胎停止发育并死亡。若同时皮下注射黄体酮1mg/只或绒毛膜促性腺激素(hCG)10IU/只均能有效地对抗半夏蛋白的抗早孕作用。但是抗早孕剂量的半夏蛋白皮下注射一次性给药并不影响早孕小鼠子宫胞浆孕酮受体的含量及亲和力。以上结果提示,半夏蛋白影响了卵巢黄体功能使内源性孕酮水平下降导致蜕膜变性,胚胎失去蜕膜的支持而流产。

EFFECTS OF PINELL1A TERN ATE BREIT PROTEIN ON THE TERMINATION OF EARLY PREGNANCY IN MICE AND STUDIES OF ITS MECHANISM

Abstract:The effects of Pinellia ternate Breit protein on early murine pregnancy were studied. The extract was given in a single 30 mg/kg dose H., while 0.9% NaCl wag given to the control animals.Twenty four hours later, plasma progesterone and oytoplasmic progesterone receptors of the murine uteri were assayed. Eight days after adminstration, the animals were sacrificed and the embryos were examined.It was found that Pinellia ternate Breit protein could effectively inhibit (100% success) early pregnancy in mice plasma progesterone was decreased, microscopic appearance of endometrium was thin, most areas were devoid of decidua and the embryos ceased growing and died. If progesterone (1mg) or hCG (10 iu) was injected S. C. simultaneously with Pinellia ternate Breit protein, the inhibitory effects of the Pinellia ternate Breit protein did not appear.However, Pinellia ternate Breit protein at 30mg/kg S. C. had no effect on the amount of cytoplasmic progesterone receptors and its affinity to progesterone in early pregnant tnurine uteri.Accordingly, it seems that the anti-early pregnancy effects of Pinellia ternate Breit protein leads to decrease of endogeneous progesterone from the corpus luteum of ovary, decidual degeneration and death of embryos.
