摘    要:目的 :研究外源性白血病抑制因子 (LIF)及其抗体对小鼠胚胎着床的影响。方法 :将妊娠 4d的小鼠胚胎种植于已建立的子宫内膜体外模型上 ,观察不同浓度的LIF及其抗体对小鼠胚胎的粘附、植入及扩展情况。并用RT PCR法测定子宫内膜上皮细胞整合素β3的变化。结果 :不同浓度的LIF促进了胚胎的粘附 (P <0 .0 5 ) ,胚胎的扩展率降低。加入LIF抗体 (5 ,1 0 μg/ml)降低了胚胎的粘附率 (P <0 .0 5 ) ,同时LIF对整合素β3的表达有明显的促进作用 (P <0 .0 1 ) ,加入抗体后整合素β3的表达降低。结论 :LIF可能通过上调整合素β3的表达促进胚胎的粘附和植入

Abstract:Objectives:To study effect of LIF and LIF antibody on embryo implantation.Methods:Using the established three dimensional endometrial model,the rate of attachment,implantation and outgrowth was investigated at different concentration of LIF and LIF antibody.The level of integrin β3 mRNA was measured with RT PCR.Results:LIF (0.1,1,10,100ng/ml) significantly enhanced the blastocyst attachment after 48h,( 64.6%,66.7%,68.8%,70.8%,58.3% in the control experiment),while the outgrowth was decreased(8%,10.4%,6%,10.4%,and 16.7% in the control experiment).Conversely,5μg/ml and 10μg/ml LIF antibody decreased the percentage of blastocyst attachment(39.6%,41.6%,and 58.3% in the control experiment).The level of integrin β3 mRNA were significantly higher in the group of LIF(2.00±0.07,2.61±0.07, 2.62±0.03, 2.27±0.08) than that in the control experiment(1.15±0.06).The level of integrin β3 mRNA decreased in the experiment of 10μg/ml and 5μg/ml LIF antibody.( 0.7597±0.56,0.728±0.04) (P<0.05).Conclusion:LIF may improve the blastocyst attachment and implantation by up regulating integrin β3 which can induced uterine receptivity.
