银杏内酯A和银杏内酯B在健康人体内的药动学的实验研究

 目的 用本实验室建立的测定血液中内酯浓度的方法，对银杏内酯A(ginkgolide A,GA)和银杏内酯B(ginkgolide B,GB)在10例健康受试者体内的药动学进行了研究。方法 10例健康受试者，静脉滴注银杏叶提取物注射液（舒血宁注射液），于给药后不同的时间取血，用气相-质谱联用法测定血药浓度，并计算GA和GB的药动学参数。结果 健康受试者静脉点滴GA和GB后的代谢均符合二房室模型，t_1/2分别为3.73，4.25 h。AUC值分别为68 305.10，19 714.69 ng·min·mL^-1。结论 GC-MS测定银杏内酯的血药浓度是测定银杏内酯的较好的方法。

Study on the pharnacokinetics of ginkgolides in 10 volunteers

OBJECTIVE To study the pharmacokinetics of ginkgolide A(GA) and ginkgolide B (GB).METHODS The Ginkgo biloba extract(GBE) injection was administered by intravenous infusion in 10 volunteers and GC-MS was used to determine the concentrations of GA and GB in plasma.RESULTS The dispositions of both GA and GB were conformed to two-compartment model.t_1/2 of GA and GB were 3.73 h and 4.25 h,respectively.The AUC of GA and GB were 68 305.10 ng·min·mL^-1 and 19 714.69 ng·min·mL^-1 respectively.CONCLUSION The GC-MS assay was sensitive for the determination of GA and GB in human blood.The pharmacokinetic parameters of GA and GB from 10 volunteers were similar to those reported.
