高住低训对大鼠骨骼肌SDH与LDH活性的影响

摘    要:目的:探讨高住低训(HiLo)对骨骼肌有氧代谢和糖酵解能力的影响.方法:7周龄健康雄性SD大鼠96只,随机分为4组:对照组(C)、低氧对照组(HC)、高住低训组(HT)和常氧训练组(NT),又依4天、7天、28天的不同取样时间,将以上每组分成3小组.建立模拟海拔4000米中等强度高住低训4周的动物模型(跑台坡度10%,跑速20米/分,1次/天,1小时/次,5次/周).用生化法分别测定不同时间点大鼠腓肠肌琥珀酸脱氢酶(Succinate dehydrogenase,SDH)和乳酸脱氢酶(Lactate dehydrogenase,LDH)的活性.结果:4~28天高住低训后,骨骼肌SDH活性显著升高(P<0.001),其在第28天时较对照组高约2倍;整个实验期LDH活性无显著变化(P>0.05).提示高住低训可提高骨骼肌有氧代谢能力,但对糖酵解供能能力的影响尚不确定.

Abstract:Objective The purpose of this study was to investigate the effect of living at high altitude and training at low altitude (HiLo) on the aerobic oxidative and glycolytic capabilities of skeletal muscle in rats. Methods Ninety six 7-wk-old male SD rats were randomly assigned to 4 groups: controls (C), hypoxic controls (HC), hypoxic training (HT) and normoxic training (NT). Each group was divided into 3 subgroups, and each subgroup was observed on the 4th day, 7th day and 28th day. A HiLo model was developed to simulate living at 4000m altitude while trained under normoxia with moderate intensity (treadmill grade 10%, speed 20m/min, one bout per day, 5 bouts per week and 1 hr per bout). Biochemical methods were employed to measure the changes in SDH and LDH in the gastrocnemii of experimental rats. Results The results showed that SDH activity increased significantly (P<0.001), as high as nearly 2 times on the 28th day in HT as compared with that in C, while the LDH activity changed insignificantly within the entire experimental period (P>0.05). Conclusions The study suggested that HiLo could promote the aerobic oxidative potential, whereas HiLo had no significant effect on glycolytic capability in gastrocnemii.
