基质金属蛋白酶与下咽癌侵袭转移的相关性研究

摘    要:目的:探讨基质金属蛋白酶2(MMP2)、基质金属蛋白酶9(MMP9)在下咽鳞状细胞癌组织中的表达与其生物学行为尤其是侵袭转移之间的关系。方法:免疫组织化学S—P法研究59例下咽癌组织标本和9例正常的下咽黏膜中MMP2、MMP9和Ki-67的表达。以Ki-67指数(labeling index)作为反映下咽鳞状细胞癌细胞的增殖活性程度的指标。结果:MMP2、MMP9阳性表达率明显增高。MMP2、MMP9在颈淋巴结转移阳性组的表达高。MMP9的表达与肿瘤的组织学分化程度呈正相关,与肿瘤的增殖活性呈正相关。结论:MMP2、MMP9与肿瘤侵袭转移的生物特性密切相关,可以作为下咽鳞状细胞癌侵袭转移的预后指标。

Abstract:Objective:To study the relationship between the expression of matrix metalloproteinase-2(MMP-2) , matrix metalloproteinase-9(MMP-9) and the invasion and metastasis of hypopharyngeal squamous cell carcinoma, to explore new approaches to evaluate and control the metastasis of hypopharyngeal squamous cell carcinoma. Methods: The expression of MMP-2,MMP-9, Ki-67 in the fifty-nine surgical specimens of hypopharyngeal squamous cell carcinoma and nine samples of normal hypopharyngeal mucosa were detected by streptavidin-peroxidase conjugated method (S-P). The Ki-67 labeling index was assessed to measure the tumor proliferation rate of the hypopharyngeal squamous cell carcinoma.Results: The positive expression rates of MMP-2 and MMP-9 with neck lymph node metastasis was significantly higher than those in tumors without neck lymph node metastasis.MMP-9 expression was associated with pathological grade and proliferation rate. Conclusion: MMP-2,MMP-9 may play a key role in metastasis and invasion of hypopharyngeal squamous cell carcinoma, and represent potential prognostic markers as well as ideal therapeutic targets for gene therapy.
