肺鳞癌患者癌组织和血液中微卫星DNA变异的检测

摘    要:目的:检测肺鳞癌患者癌组织和外周血中微卫星DNA的变异。方法:选择30例原发性肺鳞癌患者的癌组织、外周血液标本及30例肺部良性疾病患者的外周血液标本,用PCR—银染法,对其中的20个微卫星位点进行检测。结果:癌组织微卫星DNA变异的检出率较高(26%),肺鳞癌患者外周血20个位点微卫星DNA变异的检出率(20.2%)与癌组织相比差异均无统计学意义,检测出的敏感位点有D3S1067、D3S1447、D3S1611、D3S1284、D3S1110、D3S1007、D5S346、D9S1748。30例肺部良性疾病患者外周血在全部20个微卫星DNA位点上没有检测到变异。结论:肺鳞癌患者存在微卫星DNA的变异,进行外周血中敏感位点微卫星DNA变异的检测对肺鳞癌的早期诊断有重大意义。

Abstract:Aim : To investigate the microsatellite DNA variations in cancer tissues and blood samples of lung squamous carcinoma patients and its value to early diagnosis of lung squamous carcinoma. Method: PCR-silver staining method was used to detect microsatellite DNA in cancer tissues and blood from 30 patients with lung squamous carcinoma. Results : The rates of microsatellite DNA variation were higher, and these variations were also easily seen in cancer tissues as well as in blood samples. The sensitive gene sites included D3S1067, D3S1447, D3S1611, D3S1284, D3S1110, D3S1007, D5S346, D9S1748. Conclusion: Microsatellite DNA variations exist in lung squamous carcinoma. Detecting the variations of blood will be meaningful to early diagnosis of lung squamous carcinoma.
