应用基因芯片筛选果蝇七氟醚麻醉敏感性相关基因

摘    要:目的:探索吸入麻醉药敏感性相关基因,深入研究其作用机制。方法:应用基因芯片技术,以七氟醚敏感(S)和耐药(R)的果蝇品系为研究模型,野生型黑腹果蝇(H)为对照,筛选吸入麻醉药七氟醚敏感性相关的候选基因。结果:以H品系为对照,S品系检测到差异表达基因647个,R品系检测到375个。对这些基因进行功能分类发现在S品系中大多数参与原发性主动转运活性、单价无机阳离子转运活性等过程的基因都上调表达,而大多数参与糖类代谢等过程的基因都下调表达。同样在R品系中大多数参与糖类代谢等过程的基因都上调表达。其中有2个基因在S品系中上调表达而在R品系中下调表达,有8个基因在S品系中下调表达而在R品系中上调表达。结论:这10个变化相反的基因成为敏感相关性基因的可能性较大,有待进一步研究。

Abstract:Objective:To search new clues to reveal the action mechanism of inhaled anesthetics.Methods:Microarray was applied to compare differentially expressed genes from three different strains of Drosophila melanogaster,the sevoflurane-sensitive strain(S),the sevoflurane-resistant strain(R)and the wild type strain(H).Results:H strain was used as control group,there were 647 differentially expressed genes in S strain,and 375 in R strain.Functional category analysis found that in S strain major genes which involved the progresses of primary active transport activity,monovalent cation transporter activity were up-expressed,and major genes which involved the progresses of carbohydrate metabolism were down-expressed.However in R strain genes which involved the progresses of carbohydrate metabolism were up-expressed.There were two genes up-expressed in S strain but down-expressed in R strain,and eight genes down-expressed in S strain but up-expressed in R strain.Conclusion:The ten genes may be the genes related to sevoflurane sensitivity,further studies are expected.
