补骨脂素对大鼠前列腺增生治疗作用及对前列腺细胞雌雄激素受体的影响

摘    要:目的探讨补骨脂素对大鼠前列腺增生的治疗作用及对前列腺细胞雌激素受体(ER)和雄激素受体(AR)的影响.方法将100只SD大鼠随机均分为5组,除正常组外,其余各组去势,7 d后除正常组、前列腺增生组外,皮下注射丙酸睾酮,同时补骨脂素组灌胃给予补骨脂素,阴性对照组给予等量生理盐水,保列治对照组给予保列治.30 d后处死,称取前列腺湿重,应用免疫荧光流式细胞法检测前列腺ER和AR的表达.结果补骨脂素组大鼠前列腺湿重低于各对照组(P<0.05),ER和AR的标记率明显下降(P<0.05).结论补骨脂素对大鼠前列腺增生有明显治疗作用,其作用机制可能是通过抑制前列腺细胞ER和AR的表达而实现的.

Abstract:Objective To investigate the therapeutic effect of psoralen on prostatic hyperplasia and its influenceson estrogen receptor (ER) and androgen receptor (AR) of prostatic cells in SD rats. Methods One hundred SDrats were randomly divided into five groups (each n=20) and were castrated except the normal group. After 7days testosterone propionate was injected subcutaneously to the rats except the normal and prostatic hyperplasiagroups, and at the same time the psoralen group was given psoralen orally. The control group was given normalsaline solution and the Proscar group was given Proscar. The rats were killed after 30 days. The wet weight ofrat prostate was weighed and the expressions of ER and AR of prostatic cells were detected by using immunofluo-rescence flow cytometry. Result The wet weight of rat prostate in the psoralen group was lower than that of oth-er groups (P<0.05). The tagged rates of ER and AR of prostatic cells were obviously decreased (P<0.05). Con-clusion Psoralen has a remarkable curative effect on rat prostatic hyperplasia. The mechanism may be related tothat it can inhibit the expressions of ER and AR of prostatic cells.
