摘    要:【目的】探讨经鼻给予神经生长因子治疗大鼠脑梗塞后脑内巢蛋白的表达。【方法】选用SD大鼠72只,随机分为3组:模型组(n=24)、对照组(n=24)和治疗组(n=24),采用线栓法建立大鼠局灶性脑缺血模型,对照组经鼻给予等量生理盐水,治疗组于脑缺血后2h经鼻给予神经生长因子,每组又根据处死时间不同分为3、7、14、21d组,用Longa等评定方法检测大鼠行为、免疫组化方法检测脑内海马区巢蛋白的表达。【结果】神经功能评分在各时问点治疗组与模型组、对照组比较差异有统计学意义;大鼠脑缺血后各组问不同时相均有巢蛋向阳性细胞数,治疗组7d阳性细胞数量显著增加。【结论】经鼻给予神经生长因子可有效促进脑缺血后脑内巢蛋白的表达,有助于大鼠神经功能恢复。

Influence of intranasal NGF on expression of Nestin in ischemic cortex of adult rat with cerebral ischemia

Abstract:[Objective]To investigate the effect of Intranasal NGF on the expression of nestin after focal cerebral ischemia in adult rats. [ Methods ] A total of 72 SD rats were randomly divided into 3 groups: model group, control group, and treatment group. Focal cerebral ischemia models were induced by the thread occlusion method. Then each group was subdivided into 4 groups according to the time of executing the animal (3,7, 14, 21 days) , control group was intranasally giving 0.9% Nacl and Rats in experimental group were intranasally given NGF zhafter cerebral ischemic. Using Longa evaluation method to detect the behavior of rats, immunohistochemistry was used to detect the expression of uestin in the hippocampus of the hrain. [ Results]The score points in treatment group at each time difference of nerve function was significant, there were nestin positive ceils at different time points after cerebral ischemia, and the number of nestin positive cells in treatment group were increased significantly on the 7th day. [Conclusion] Intranasal NGF can enhance the expression of nestin positive ceils in some brain regions after focal cerebral ischemia, contribute to the recovery of nerve function.
