氧化铝固相萃取小柱在苦参生物碱含量测定中的应用

摘    要:该文使用中性氧化铝Cleanert Alumina-N-SPE小柱(0.5 g/6 mL),洗脱剂为三氯甲烷-甲醇(7:3)5 mL,代替氧化铝开放柱(100~200目,5 g,内径1 cm),依次以三氯甲烷、三氯甲烷-甲醇(7:3)各20 mL进行洗脱对苦参药材供试液进行处理,该方法优化洗脱溶剂使用种类和数量,并优化了苦参药材中苦参碱和氧化苦参碱含量测定的色谱条件,将2010年版《中国药典》中流动相条件乙腈-无水乙醇-3%磷酸溶液(80:10:10)优化为乙腈-无水乙醇-3%磷酸溶液(84:6:10);简化了2010年版《中国药典》规定苦参药材供试液的前处理,缩短了样品处理时间,减少了洗脱溶剂,结果更为准确可靠,适合苦参药材中含量的测定,提高苦参药材中苦参生物碱含量测定的工作效率。

Abstract:A Cleanert Alumina-N-SPE column (0.5 g/6 mL) chromatograpy with 5 mL of chloroform-methanol (7:3) as eluent,instead of aluminum oxide column (100-200 mesh, 5 g, l cm) chromatograpy eluted successively with chloroform and the chloroform-methanol (7:3)(20 mL each), was applied to enrich matrine and oxymatrine in Sophora flavescens. Also, the optimization of the HPLC determination conditions with acetonitrile-ethanol absolute-3% phosphoric acid solution (84:6:10) as mobile phase, instead of acetonitrile-ethanol absolute -3% Phosphoric acid solution (80:10:10) recorded in Chinese Pharmacopoeia 2010 Edition, was more suitable for determination of matrine and oxymatrine in S. flavescens. This method has advantage of reducing sample handling time and solvent volume and increasing the accuracy and feasibility, which can simplify the procedure for determination of matrine and oxymatrine in S. flavescens.
