摘    要:

目的  探讨白藜芦醇对大鼠脑出血后血肿周围组织细胞凋亡的影响及其作用机制。方法  采用二次注血法制作脑出血模型。将大鼠随机分为治疗组、对照组和假手术组,每组分3、6、12、24、48、72和168 h共7个时间,每个时间6只大鼠。利用蛋白印迹法和免疫组织化学法对P-JAK2、P-STAT3、Bcl-XL、Caspase-3蛋白做定量和定性分析。利用原位末端标记法检测细胞凋亡指数的变化。结果  治疗组在各个时间细胞凋亡指数低于对照组(P <0.05),假手术组中细胞凋亡指数低于对照组与治疗组(P <0.05)。治疗组在各个时间的P-JAK2、P-STAT3、Bcl-XL蛋白的表达高于对照组(P <0.05),Caspase-3蛋白的表达低于对照组(P <0.05),假手术组中4种蛋白表达低于对照组与治疗组(P <0.05)。结论  白藜芦醇可以促进JAK2、STAT3的磷酸化,增加Bcl-XL表达,降低Caspase-3表达,有抗细胞凋亡的作用。

Abstract:

Objective To investigate the effect of resveratrol on apoptosis of tissue around hematoma and its mechanism in rats with intracerebral hemorrhage. Methods The cerebral hemorrhage model was made by the two-step injection of blood. The rats were randomly divided into three groups including treatment group, control group and sham-operation group. Each group was subdivided into 3-, 6-, 12-, 24-, 48-, 72- and 168-h groups with 6 rats in each group. The levels of p-JAK2, Bcl-xl, p-Stat3 and caspase-3 proteins were quantitatively and qualitatively analyzed by Western blot and immunohistochemistry. TUNEL method was used to detect the change of apoptosis index. Results The apoptosis index of the treatment group was lower than that of the control group at each time point (P < 0.05). The apoptosis index in the sham-operation group was lower than that of the control group and the treatment group (P < 0.05). The expressions of p-JAK2, p-Stat3 and Bcl-xl proteins were higher in the treatment group than in the control group at each time point (P < 0.05) whereas the expression of caspase-3 protein was lower than that of the control group (P < 0.05). The expressions of the four proteins in the sham-operation group were lower than those in the control group andthe treatment group (P < 0.05). Conclusions The anti-apoptosis effect of resveratrol is to promote the phosphorylation of Stat3 and JAK2, and thus to increase the expression of Bcl-xl and reduce the expression of caspase-3.
