摘    要:目的研究大鼠急性肺栓塞模型肺组织中血管内皮生长因子(VEGF)、缺氧诱导因子1(HIF-1α)的表达变化,以了解阿E与VEGF、HIF-1α相关性。方法建立大鼠急性肺栓塞模型,分别在急性肺栓塞后1、6、12、24h开胸取肺组织。常规提取肺组织的总蛋白,以正常组为对照,免疫组织化学法测定VEGF、HIF-1α表达水平的变化。结果在大鼠急性肺栓塞后,HIF-1和VEGF蛋白水平均逐渐升高,在6h升高最为明显。结论HIF-1α在大鼠PTE肺组织中呈高表达,并调节其下游基因VEGF的表达增强,可对肺组织产生保护作用。

Abstract:Objective To study the changes of VEGF and HIF-1α expression in a rat model of acute pulmonary embolism (PE) and their correction. Methods Lung tissue samples were collected 1, 6, 12 and 24h after a rat model of acute PE was established. Total protein was extracted from the lung tissue samples. Normal rats were used as controls. Immunohistochemical method was used to examine changes of HIF-1α and VEGF expression in lung tissue samples. Results The VEGF α nd HIF-1α expression levels were gradually increased after acute PE and reached their peak at 6h after acute PE. Conclusion The expression of HIF-1 is up-regulated in rat lung tissue after acute PE and increases the expression of VEGF, thus protecting lung tissue from damage.
