摘    要:目的:探讨替米沙坦对自发性高血压大鼠(SHR)血管前纤维蛋白1(Profmn-1)及细胞外信号调节激酶(ERK1/2)磷酸化水平的影响。方法:选取10周龄SHR及其同源对照魏-凯氏(WKY)大鼠,给予替米沙坦(5~10mg/kg·d)或安慰剂,为期10周。尾套法测量大鼠尾血压。采用实时定量聚合酶链式反应(PCR)和Western免疫印迹检测治疗后大鼠主动脉组织中Profilin-1mRNA和蛋白及ERK1/2磷酸化水平。结果:与WKY对照组相比,SHR大鼠血压明显升高(126±3.5)mmHg:(195±6.1)mmHg],伴血管Profilin—1mRNA(1.0±0.11);(7.8±0.57)]及ERK硫酸化水平(1.0±0.11):(2.43±0.19)]表达明显升高(P均〈0.01),而经替米沙坦治疗后SHR大鼠血压明显降低替米沙坦低剂量组(169±6.2)mmHg,高剂量组(161±4.9)mmHg],伴Profilin-1mRNA替米沙坦低剂量组(4.45±0.92),高剂量组(1.95±0.41)]表达及ERK1/2磷酸化水平替米沙坦低剂量组(1.62±0.20),高剂量组(1.34±0.09)]明显下调(P均〈0.05)。结论:长期替米沙坦治疗可降低高血压大鼠血压水平,降低血管Profilin-1表达及ERK1/2磷酸化水平,提示替米沙坦对高血压血管重塑具有一定的保护功效。

Abstract:Objective: To investigate the effects of telmisartan on vascular expression of Profilin-1 and extraeellular regulated kinase (ERK1/2) in spontaneous hypertension rats (SHR). Methods.. The 10 weeks SHR and Wistar- Kyoto (WKY) rats were treated with telmisartan (5 or 10 mg/kg ·d) or placebo for 10 weeks. Systolic blood pressure (SBP) of rat was measured by tail-cuff method. The mRNA, protein and phosphorylation levels of Profilin-1 and ERK1/2 in treated rats were determined with real-time polymerase chain reaction, Western blotting methods respectively. Results.. Compared with WKY rat, the SBP significantly elevated (126± 3.5) mmHg; (195±6.1) mmHg], accompanied by enhanced Profilin-1 mRNA (1.0±0.11) : (7.8±0. 57)] and phosphorylated ERK1/2 (1.0±0.11) ; (2.43±0.19)] levels in SHR (P〈0. 01 all). Treatment with Telmisartan significantly reduced the SBP Telmisartan low dose group (169±6.2) mmHg, high dose group (161±4. 9) mmHg], along with reductions in Profilin-1 mRNA expression -Telmisartan low dose group (4.45±0. 92), high dose group (1.95±0.41)] and ERK1/2 phosphorylation levels Telmisartan low dose group (1.62±0.20), high dose group (1.34±0. 09)], P〈 0.05 all. Conclusion: Chronic Telmisartan treatment reduces systolic blood pressure and down-regulates vascular expression of Profilin-1 and ERK phosphorylation levels in SHR, suggesting that Telmisartan have potential vascular protective effects in hypertension.
