醋酸棉酚对人腺样囊性癌ACC-M细胞生长及E-cadherin基因甲基化的影响

摘    要:目的:研究醋酸棉酚(gossypol acetic acid,GAA)对体外培养的人腺样囊性癌ACC—M细胞生长及E—cadherin基因甲基化的影响。方法:应用四唑盐比色法(MTT)观察GAA对人腺样囊性癌ACC—M细胞生长的作用。应用甲基化特异性PCR(MS-PCR)检测ACC—M细胞在GAA作用48、72h后E-cadherin基因甲基化状态的改变情况。结果:MTT法检测显示:GAA作用ACC—M细胞24、48和72h后,细胞的生长受到抑制。MS—PCR检测结果显示:分别以19、10μmol/L的GAA作用ACC—M细胞48、72h后,E—cadherin甲基化条带的平均灰度值低于对照组(P〈0.05)。结论:GAA能抑制ACC—M细胞生长,降低E—cadherin基因的甲基化水平,这可能是GAA抗肿瘤作用的机制之一。

Effects of Gossypol Acetic Acid on Proliferation and DNA Methylation Level of E--cadherin Gene in Human Adenoid Cystic Carcinoma ACC--M Cell

Abstract:Objective: To study the effects of gossypol acetic acid (GAA) on proliferation and methylation level of E--cadherin in human adenoid cystic carcinoma ACC--M cell in vitro. Methods: MTT assay was used to detect the effects on proliferation in ACC--M cells treated by different concentrations of GAA. Methylation specific PCR (MS --PCR) was used to detect methylation level changing of E--cadherin gene in ACC--M ceils treated by GAA for 48h and 72h. Results: Experimental GAA group could be observed on growth and proliferation inhibition of E--cad- herin cells after being treated by GAA. Compared with the control group, MS--PCR showed the hypermethylation of E--cadherin gene was partly reversed after GAA treatment (19μmol/L GAA for 48h and 10μmol/L for 72h)(P〈0.05). Conclusion: GAA can suppress proliferation of ACC--M ceils and has demethylation effect on its E--cad- herin gene, which may be one of its tumor suppression mechanisms.
