大鼠角膜创伤愈合过程中HSP70表达的实验研究

摘    要:目的 观察大鼠角膜创伤愈合过程中热休克蛋白70(HSP70)的表达规律,探讨其在创伤愈合中的作用.方法 雌性Wistar大鼠42只,随机分为7组,每组6只,其中,正常对照组6只,动物模型组36只.均取右眼制作角膜针刺损伤模型,于损伤后各时间点(6h,1d,3d,5d,7d,14d)取角膜标本,HE染色行组织病理学检查,免疫组化染色观察HSP70在大鼠角膜的表达与分布,用计算机图像系统对结果进行分析.结果 HSP70在大鼠正常角膜各层均有表达,动物模型组大鼠角膜上皮HSP70的表达水平于损伤后第1天显著增高(P〈0.05),其后逐渐下降,至损伤后第14天时恢复正常.角膜基质层和内皮层HSP70的表达在各时间点未见明显变化.结论 HSP70在大鼠角膜创伤愈合过程中的早期表达水平明显增高,其可能是参与调节角膜创伤愈合的一种重要分子.

Abstract:Objective To observe the expression of heat shock protein 70( HSP70 ) during corneal wotmd healing in rats, so as to explore its effeet on wound healing. Methods Forty-two female Wistar rats were divided into 7 groups randomly,6 rats in each group. There were 6 rats in normal corneal group,and 36 in injury model groups. The right eyes were used to establish corneal injury models by needle puncture. The corneas were surgically removed on six different time points after the injury,that was 6h, ld,3d,5d,7d and l4d. HE stain was used for histopathulogical examination and immunohistoehemical stain was used to observe the expression and distribution of HSP70 in the corneas. The computer image system was used to analyze the results. Results HSP70 was expressed in the normal cornea. The expression level of HSP70 of injury groups reached the maximmn on the 1st day in the corneal epithelium after the injury,the difference was significant compared with normal corneal group( P 〈0. 05) ,then decreased gradually,and it reached to nomal level on the 14th day. The expression of HSP70 did not change obviously on any time point in corneal stroma and endothelial layer. Conclusion The expression level of HSP70 increased signifieantly during the early period of corneal wound healing in rats. HSP70 maybe an important molecule involved in the regulation of corneal wound healing.
