吉非替尼联合5-氟尿嘧啶对HER-1高表达胃癌细胞的体外实验研究

目的初步探讨吉非替尼（Iressa）联合5-FU对HER1高表达胃癌细胞的生长抑制作用。方法以Her-1基因高表达的胃癌细胞株SGC7901为研究对象,以MTT法检测两药物各自及合用时对胃癌细胞株增殖影响的抑制率;流式细胞仪检测细胞周期变化和凋亡;以RT-PCR分析药物对靶蛋白酶mRNA的影响。结果吉非替尼可增强化疗药对HER-1阳性胃癌细胞的毒性作用。先吉非替尼和5-FU联合再序贯使用分子靶向药吉非替尼的方案效果最佳;联合方案对细胞周期的改变影响明显;同时吉非替尼能增强5-FU对胸苷酸合成酶（TSmRNA）表达的抑制。结论吉非替尼联合5-FU对Her-1/EGFR过表达胃癌细胞有明显的杀伤性增强作用而且存在最佳联合方案。

The Experimental Study Of Gefitinib Combined With 5-FU On Over-expressed HER-1 Gastric Cancer Cell In Vitro

Objectives To assess the growth inhibition of Gefitinib(Iressa) in combination with 5-fluorouracil(5-FU) on over-expressed HER-1 gastric cancer cell lines.Methods Gastric cancer cell lines SGC7901 of her-1 gene high expression was the research object,Detecting the inhibitory effects of gifitinib,chemotherapy drug alone or in combination on gastric cancer cells by MTT assay;Flow cytometry was used to determine the effect of gifitinib and 5-FU alone or in combination on cell cycle and on the apoptosis of human gastric cancer cells;Detecting the drug effect on target protease mRNA by RT-PCR method.Results The effect of chemotherapy drugs’ cytotoxicity on over-expressed HER-1 gastric cancer cells was augmented by gifitinib.The best scheme was gifitinib combined with 5-FU and then sequential used targeted molecular medicine alone;A significant change appearred in the cell cycle when gifitinib in combination with 5-FU;The inhibition of 5-FU on TSmRNA can be enhanced by gifitinib.Conclusion Gifitinib in combination with 5-FU on over-expressed HER-1 gastric cancer cells in vitro can increase cytotoxicity and exist the optimal schedule.
