银杏叶提取物对结肠癌SW480细胞增殖的影响

目的研究银杏叶提取物（EGB）对SW480结肠癌细胞生长增殖的影响。方法体外培养人结肠癌SW480细胞后,给予不同浓度（100,200,300,400,500 mg·L^-1）银杏叶提取物干预后,采用CCK-8检测人结肠癌SW480细胞的增殖抑制率,选择最佳作用浓度;随后,在此作用浓度下观察EGB作用不同时间对人结肠癌SW480细胞增殖抑制率的影响,并通过Western blot检测人结肠癌SW480细胞内Ki67和PCNA的表达情况。结果银杏叶提取物可抑制结肠癌SW480细胞的增殖,且随着作用浓度增加和作用时间的延长其抑制作用逐渐增强;300 mg·L^-1银杏叶提取物对结肠癌SW480细胞增殖的抑制效果最明显。银杏叶提取物干预的结肠癌SW480细胞内Ki67和PCNA的表达均明显下调。结论银杏叶提取物可呈浓度和时间依赖性地抑制结肠癌SW480细胞的增殖。

The Effects of Oral Ginkgo Biloba Extract (EGB) on the Proliferation of Human Colon Cancer SW480 Cell Lines

Objective To investigate the effects of oral ginkgo biloba extract（EGB） on the proliferation of SW480 colon cancer cell lines. Methods SW480 colon cancer cell lines were cultured in vitro. CCK-8 assay was used to measure the inhibition rate of SW480 proliferation after treated by 100, 200, 300, 400 and 500 mg·L^-1 EGB. Selected out the optimize concentration of EGB and observed its influencing difference when treating the SW480 cell for different time. The expression of proliferation related indicators Ki67 and PCNA was detected using western blot. Results EGB inhibited the proliferation of SW480 cells with the increase of EGB concentration and treatment time. Both the expressions of Ki67 and PCNA decreased after GBM intervention. Conclusion EGB significantly suppressed the proliferation of colon cancer cell lines SW480 in a concentration and time dependent manner.
