Netrin-1在肝癌组织中的表达及其对肝癌细胞侵袭能力的影响

目的 观察神经轴突导向因子Netrin-1在人肝细胞癌(HCC)组织中的表达及其对肝癌细胞侵袭能力的影响.方法 应用免疫组织化学SP方法检测45例肝癌手术标本癌旁及癌组织中Netrin-1的表达水平,用卡方检验分析其与临床病理参数的关系.在肝癌细胞SMMC-7721中转染Netrin-1基因,观测过表达Netrin-1肝癌细胞侵袭能力的改变.结果 Netrin-1在肝癌中的阳性表达率约为66.7%,主要在肝癌细胞胞质中表达,Netrin-1的阳性表达与性别、年龄、肿瘤大小以及细胞分化程度无关(P>0.05),『fii与门静脉痛栓以及淋巴转移有关(P<0.05).细胞侵袭实验结果显示,与实验对照组比较过表达Netrin-1显著促进肝癌细胞SMMC-7721侵袭能力(P<0.05).结论 HCC组织中高表达的Netrin-1与肿瘤侵袭转移有关,Netrin-1促进肝癌细胞侵袭能力可能是肝癌转移机制之一.

Expression of Netrin-1 in hepatocellular carcinoma tissue and its effect on the invasion of hepatocellular carcinoma cells

Objective To study Netrin-1 expression in hepatocellular carcinoma tissue and its effect on invasion of hepatocellular carcinoma cells. Methods The expression of Netrin-1 in 45 cases of hepatocellular carcinoma was detected by immunohistochemistry. The correlation between Netrin-1 and clinicopathologic features was analyzed by Chi-Squared test. The effect of Netrin-1 on invasion of SMMC- 7721 ceils was analyzed by invasion assay. Results The positive rate of Netrin-1 in HCC tissue was 66.7%. Netrin-1 expression was mainly located in kytoplasm of liver cancer cells. Netrin-1 expression was associated with lymph node metastasis, portal vein invasion, and metastatic lesions(P < 0.05 ), bet not with sex, age, tumor size, and cell differentiation (P >0.05 ). Cell invasion assay revealed that the number of migrating SMMC-7721 cells induced by Netrin-1 was increased(P < 0.05). Conclusion The up-regu- lation of Netrin-1 in HCC has a close relationship to the invasion of hepatoma cells. Invasion of hepatoma cells induced by Netrin-1 may be one of mechanisms of metastasis of HCC.
