耐力训练及补充中药多糖对大鼠PBMC分泌细胞因子和血清中细胞因子含量的影响

摘    要:目的:探讨6周耐力训练及补充多糖提取物对大鼠血清中3种细胞因子水平和PBMC分泌3种细胞因子能力的影响。方法:120只雄性Wistar大鼠随机分为6组:耐力训练+黄芪多糖组、耐力训练+牛膝多糖组、单纯耐力训练组、安静+黄芪多糖组、安静+牛膝多糖组、安静对照组。大鼠进行6周递增负荷游泳训练后,观察大鼠血清中IL-2、IL-4、IL-10水平的改变以及外周血单个核淋巴细胞(PBMC)分泌IL-2、IL-4、IL-10能力的变化。结果-K期耐力训练后可造成大鼠血清IL-2下降、IL-4、IL-10升高,PBMC分泌IL-4能力增高,表明长期耐力训练后Th1和Th2两型细胞的功能失衡。6周耐力训练同时补充黄芪多糖和牛膝多糖提取物能防止大鼠血清IL-2的明显下降及IL-4、IL-10升高;防止大鼠PBMC分泌IL-4的明显增高。结论:长期耐力训练能引起辅助T细胞(Th)两个亚型Th1和Th2两型细胞功能失衡,这可能是长期耐力训练引起运动性免疫低下的原因之一。牛膝多糖和黄芪多糖提取物对耐力训练大鼠Th1和Th2型细胞平衡有一定的调节作用。

Abstract:Objective:Study of intensive endurance training and supplementation of herbal polysaccharide extracts on levels of IL-2, IL-4,IL-10 in serum of rats.Methods:120 Wistar Rats were randomly divided into 6 groups: sedentary control group(n=20), sedentary with supplementation of APS group(n=20), sedentary with supplementation of ABPS group(n=20), training group(n=20), training with supplementation of APS group(n=20) and training with supplementation of ABPS group(n=20). After 4-week and 6-week swimming, IL-2, IL-4,IL-10 in serum of rats and supernatant of PBMC were tested through ELISA.Results:After 6 weeks training, (1) In training group, the level of IL-2 in serum was lower than that in sedentary control group(P<0.05). On the contrary, the levels of IL-4,IL-10 in serum were increased evidently(P<0.05), while the level of IL-4 in the supernatant of PBMC was increased. (2) The changes of IL-2, IL-4 and IL-10 were inhibited by supplementation with ABPS or APS.Conclusion:The results indicate that supplementation with ABPS or APS may help regulate the balance of Th1 and Th2 through modulating the levels of IL-2 , IL-4 and IL-10 in rats during strenuous training.
