水通道蛋白1、3在膀胱尿路上皮癌中的表达及临床意义

摘    要:目的探讨膀胱尿路上皮癌中水通道蛋白1、3(AQP1、AQP3)的表达与分布,及其与组织学分级和病理分期之间的关系及临床意义。方法采用免疫组化S-P法、蛋白免疫印迹(Western-blot)法对60例膀胱尿路上皮癌组织及30例正常膀胱组织中AQP1及AQP3的表达进行检测,并分析与其不同组织学分级、不同病理分期和有无淋巴结转移的关系。结果 AQP1在膀胱尿路上皮癌组织及正常膀胱组织中主要表达在微血管的内皮细胞的细胞膜及细胞质中,AQP3主要表达在膀胱尿路上皮癌的癌细胞的胞膜及胞质中。免疫组织化学S-P法和Western blot法检测的结果基本一致。正常膀胱组织与不同分级的膀胱尿路上皮癌组织中AQP1及AQP3的蛋白表达量分别比较,差异均有统计学意义(P<0.01);正常膀胱组织与不同病理分期的膀胱尿路上皮癌组织中AQP1及AQP3的蛋白表达量比较,差异均有统计学意义(P<0.01);AQP1及AQP3在有淋巴结转移的膀胱癌组织中的表达量分别与无淋巴结转移的膀胱癌组织比较,差异均有统计学意义(P<0.01)。结论 AQP1和AQP3的表达与膀胱癌的组织学分级及病理分期有关,在膀胱癌形成及发展过程中起重要作用,对预测膀胱癌的侵袭性、分期及临床诊疗有指导意义。

Expressions and Significance of AQP1 and AQP3 in Bladder Urothelial Carcinoma Tissues

Abstract:Objective To investigate the expression,distribution and significance of aquaporin 1(AQP1)and aquaporin 3(AQP3)in the bladder urothelial carcinoma tissues and to investigate its relationship with histological grade and pathological stage.Methods Immunohistochemistry S-P method and Western-blot method were used to detect the expression of AQP1 and AQP3 in 60 patients with bladder urothelial carcinoma and 30 patients with normal bladder tissues.The relationship between their expressed,histological grades,pathological phases and lymphatic metastasis was analyzed.Results The AQP1 of the bladder urothelial carcinoma and normal bladder tissues mainly had expressed at membrane and cytoplasm of microvascular endothelial cells,while AQP3 mainly had expressed at membrane and cytoplasm of the carcinoma cells of the bladder urothelial carcinoma.Immunohistochemistry S-P method and Western-blot method showed coherent results.The protein expressions of AQP1 and AQP3 in normal bladder tissues and bladder urothelial carcinoma of different grades showed statistically significant difference(P<0.01).The protein expressions of AQP1 and AQP3 in normal bladder tissues and bladder urothelial carcinoma of different pathological phases showed statistically significant difference(P<0.01).The expressions of AQP1 and AQP3 in bladder urothelial carcinoma with lymphatic metastasis and in bladder urothelial carcinoma without lymphatic metastasis showed statistically significant difference(P<0.01).Conclusion The expressions of the AQP1 and AQP3 are connected with histological grade and pathological staging of bladder cancer,and it plays an important role in the formation and development of bladder cancer.It is of instructive significance in the prediction of the invasive,staging and clinical diagnosis and treatment of bladder cancer.
