单核细胞趋化蛋白-1-2518A/G基因多态性与急性心肌梗死的相关性研究

摘    要:目的研究辽西地区汉族人群中单核细胞趋化蛋白-1(MCP-1)-2518A/G基因多态性与急性心肌梗死(AMI)的相关性。方法收集2010年9月—2011年6月于辽宁医学院附属第一医院心内科住院的无血缘关系的150例AMI患者(病例组)和同期159例健康体检者(对照组)全血。采用聚合酶链反应限制性片段长度多态性(PCR-RFLP)技术检测MCP-1的基因型;ELISA法检测AMI患者和健康对照者血清MCP-1的水平。结果 AA、AG、GG 3种基因型在病例组和对照组间分布差异有统计学意义(P<0.01)。等位基因A、G的频率在病例组和对照组间分布差异有统计学意义(P<0.01);血清MCP-1水平在病例组与对照组间、病例组内不同基因型间的分布差异均有统计学意义(P<0.01)。结论 MCP-1-2518A/G基因多态性与AMI的发病具有相关性,G等位基因可能是辽西地区人群AMI的遗传易感基因,但这种联系可能和传统危险因素协同存在。

Abstract:Objective To investigate the correlation of MCP-1-2518 A/G gene polymorphism with acute myocardial infarction(AMI) in Chinese Han population in Liaoxi Province.Methods Totally 150 unrelated AMI inpatients in our department from September 2010 to June 2011 were selected as case group.Meanwhile,159 healthy persons(confirmed by physical check-ups) during the same period were recruited as the controls.MCP-1 genotype was detected by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP).The serum level of MCP-1 was determined by enzyme linked immunosorbent assay(ELISA).Results There was a significant difference in genotype distribution of three types of genes(AA,AG,and GG) between these two groups(P<0.01).The frequency distribution of A allele was significantly different from G allele(P<0.01).The distribution differences of serum level of MCP-1 between the two groups,and between different genotypes in the case group were all significant(P<0.01).Conclusion The MCP-1-2518A/G gene polymorphism is associated with the pathogenesis of AMI,and G allele may be a genetic risk factor for AMI,although such relation may co-exist with the conventional risk factors.
