摘    要:顺铂是治疗卵巢癌最有效的药物之一。为了进一步明确腹腔给药扔优越性,本实验通过腹腔与静脉药比较顺狂在小鼠腹水瘤细胞DNA中的含量变化,为临床选择用药途径提供理论依据。方法:将雌性纯系BALBC/C小鼠60只,随机分6组,每组10只,其中3组分别经腹腔给药,另3组分别经静脉给药,应用原子吸收光谱法测定荷肝癌腹水型小鼠腹腔或静脉顺铂给药后不同时间占与DNA结合的总铂量。

The influence of different route of administration of cisplatinon binding-platinum the content of DNA in mice ascitic hepatoma cells
Abstract:Purpose: Cisplatinum is one of most effective drug in combination chemotherapy of ovarian cancer. In order to confirm the superiority of IP route in the clinical experiment compared the content of DNA binding-platinum in mice ascitic hepatoma cells after IP or IV route of administration. Method: Sixty inhered Balbc/c mice were divided into six groups at random. there were ten mice in each group, in three groups of them were given cisplatinum (DDP) by IP and the other 3 groups by IV respectively. After IP and IV administration with a dose 30 mg/kg DDP, The content of DNA binding-platinum of mice ascitic hepatoma cells was determined respectively using flameless atomic absorption spectrometry. Result: The experiment showed that the total binding-platinum content was not different at 4 h after IP or IV adminstration (P>0.05). But the total binding-platinum contents at 24 and 48 h after administration were markedly different (P<0.01). Total binding-platinum contents of IP administration was higher than that of IV administration. Content of binding-platinum of IV administration declined rapidly. In contrast, content of DNA binding-platinum of IP administration declined slowly. Content of DNA binding-platinum at various time point was not different markedly. Conclusions: These data suggested that IP administration of cisplatin could enhance total platinum content binding to DNA of hepatoma cells and would increase cytotoxicity of cisplatin against ascitic hepatoma cells.
