T细胞疫苗抗同种移植排斥反应的研究

应用Ｔ细胞疫苗于同种心脏移植的研究，用Ｃｏｎ Ａ和ＣＤ３单抗诱导受同种特异性抗原（Ｃ５７ＢＬ／６（Ｈ－２＾ｂ）小鼠脾细胞）免疫的ＢＡＬＢ／Ｃ（Ｈ－２＾ｄ）小鼠，取后者脾细胞所制备的Ｔ细胞疫苗具有延长同种心脏移植物的存活期。受Ｔ细胞疫苗接种的ＢＡＬＢ／Ｃ（Ｈ－２＾ｄ）鼠脾细胞对经Ｃｏｎ Ａ激活或未激活的Ｃ５７ＢＬ／６小鼠脾细胞皆表现出特异的强烈的增殖反应，而对同系脾细胞呈低的反应。Ｔ细胞疫苗表型分析

THE RESEARCH OF T CELL VACCINATION AGAINST ALLOGRAFT REJECTION REACTION

We investingated the role of T cell vaccination on the allogous heart transplantation.T cell vaccination was made from the spleen cells of BALB/C(H 2 d)mice, which were induced by Con A or CD3 McAb and were challenged with the allologous specific antigen (C57BL/6(H 2 b) Spleen cell). The vaccination could prolong the survival of allologous heart transplant. The proliferative response of spleen cells of BALB/C(H 2 d) Challenged by T cell vaccination was assayed by Con A induced or non induced spleen cells of C57BL/6(H 2 b) mice. Both responses were very strong, but on homologous spleen cells the response was low. The analysis of phenotypes on T cell vaccination showed that the number of CD8 + clones induced by Con A was more than CD3 McAb. This suggested that T cell vaccination generate antiidiotypic T cell recognizing allologous specific antigen. It could modulate host versus graft reaction.
