头孢克肟分散片溶出度HPLC法与UV分光光度法测定结果的比较

摘    要:目的:比较HPLC法与UV分光光度法测定头孢克肟分散片的溶出度值,并从中选择适宜的方法。方法:HPLC法采用KromasilC18色谱柱,流动相为10%四丁基氢氧化铵溶液一乙腈(72:28),流速为1mL/min,检测波长为254nlyl,柱温为40℃,进样量为20μL;UV分光光度法采用溶出介质为pH7.2磷酸盐缓;中液,检测波长为288nm,浆法,转速为50r/min。结果:2种测定方法测得的头孢克肟浓度在10.60~212.10μg/mL范围内均呈线性关系(r2=1)。结论:两种方法测得头孢克肟溶出度的结果基本一致。

Abstract:Objective: To compare the dissolution of cefixime dispersible tablets by HPLC and UV methods, and the select an appropriate method. Methods: HPLC method: the determination was used Kromasil C18 column, 10% tet- rabuty lammonium hydroxide-acetonitrile(72:28) as the mobile phase at the flow rate of 1 mL/min.The detective wave- length was 254 nln, the column temperature was 40 ℃ and the injective volume was 20 μL. UV method: phosphate buf- fer(pH 7.2) was used as dissolution medium and the detection wavelength was at 288 nm. Apparatus was operated with the paddle method at the rate of 50 r/min. Results: A good linear relationship was obtained when the cefixime's concen- tration range is between 10.60 and 212.10 μg/mL(r2=l ). Conclusion: Two methods are equivalent in determination of the dissolution of cefixime dispersible tablets.
