摘    要:目的:研究化合物E-2-(3',5'-二甲氧基苯亚甲基)-环己酮(IV8)对人类宫颈癌(HeLa)、肝癌(HepG2)、前列腺癌(Dul45)和胃癌(SGC7901)细胞系的抗肿瘤活性。方法:采用MTT法评价IV8对上述4株肿瘤细胞株增殖的影响,并在光学显微镜下观察lV8对HepG2细胞形态学上的变化,以及采用AO/EB双荧光染色法评价IV8对HepG2细胞的凋亡作用。结果:IV8抑制HeLa,HepG2,Dul45和SGC7901细胞的IC。值分别为2.10,11.32,36.50和89.53gmol/L;光学显微镜下观察IV8处理的细胞出现明显增殖抑制和形态学改变;AO/EB双荧光染色IV8组可见凋亡细胞。结论:药理实验结果表明IV8具有较好的体外抗肿瘤的活性,值得进一步研究。

Abstract:Objective: To study the anti-tumor effects of 2-(3',5'-dimethoxybenzylidene)-cyclohexanone(IV8) on human cancer cell lines (HeLa, Hep O2, Dul45 and SGC7901). Methods: Compound IV8 was evaluated for antiprolif- erativc activities on the above cancer cell lines in vitro by MTT assay. The morphology alteration of Hep G2 was ob- served under inverted microscope and under AO/EB double staining. Results: Compound IV8 possessed significant in- hibito~ activity against HeLa, Hep G2, Du145 and SGC7901 with IC~ values of 2.10, 11.32,36.50 and 89.53 ~mol/L, respectively. Under the microscope, Hep G2 cells produced morphology alteration and cell number decreased signifi- cantly. AO/EB double staining found that IV8 group produced apoptotic cells.Conclusion: Preliminary pharmacological tests showed that IV8 had potent anti-tumor activities, which suggested that it was worthy of further studies.
