缺锌对大鼠肺组织IFN—r及IL—4的影响

摘    要:目的:探讨缺锌对大鼠肺组织γ干扰素(IFN—r)和白细胞介素4(IL—4)的影响。方法:SD大鼠24只,按体重随机分为3组,每组8只。A组为缺锌饲料组;B组为正常锌饲料组;C组为正常锌饲料配对饲养组。建立缺锌模型,利用酶联免疫吸附法(enzyme linked immunosorbent assay,ELISA)检测肺组织匀浆中IFN—r和IL—4的含量;RT—PCR检测肺组织中IFN—r、IL—4 mRNA表达。结果:与B组相比,A组大鼠肺组织匀浆中IFN—r含量、IFN—r mRNA的表达均明显减少(P<0.05),而IL—4含量I、L—4 mRNA的表达两组无显著差异(P>0.05)。B组与C组相比所有检测结果无显著性差异。结论:大鼠缺锌时其肺组织IFN—r明显减少,而IL—4不受影响,Thl/Th2细胞比例失衡,这可能是缺锌导致呼吸道疾病增加的重要原因。

Abstract:Objective: To investigate the effects of zinc deficiency on the expressions of interferon—γ(IFN—r) and interleukin—4(IL—4)in rats lung.Methods: Twenty-four SD rats were divided into three groups according to weight: zinc deficient diet group(group A),zinc normal diet group(group B),zinc normal pair-fed diet group(group C).Animal models of zinc-deficiency were established.The contents of IFN—r and IL—4 in lung homogenate were detected by enzyme linked immunosorbent assay(ELISA).The expression levels of IFN—r and IL—4 mRNAs were observed by RT—PCR.Results: Compared with group B,the content of IFN—r and the expression of IFN—r mRNA in lung in group A all decreased significantly(P<0.05),but the content of IL—4 and the expression of IL—4 mRNA had no difference(P>0.05).No marked difference was found between group B and group C in all results.Conclusion: Zinc deficiency reduced the expression of IFN—r but had no influence on IL—4.This imbalance of Th1/Th2 cytokine may be associated with the increasing airway diseases in zinc deficient patients.
