罗格列酮对同型半胱氨酸诱导大鼠肾脏微血管内皮细胞MCP-1和ICAM-1表达的影响

摘    要:目的了解同型半胱氨酸(Hcy)对肾脏微血管内皮细胞(RMECs)、单核细胞趋化蛋白1(MCP-1)和细胞间黏附分子1(ICAM-1)表达的影响及罗格列酮的干预作用。方法将原代培养的大鼠RMECs分为空白对照组、Hcy刺激组和罗格列酮干预组。分别采用免疫荧光法和酶联免疫吸附试验检测RMECs表达MCP-1、ICAM-1的能力。结果不同浓度的Hcy刺激后,ICAM-1的水平显著增加(P<0.05),且呈时间和剂量依赖性,尤其在0.1 mmol/L Hcy刺激24~48 h时达高峰。罗格列酮干预后ICAM-1含量显著降低,且随罗格列酮浓度的增加其抑制作用更加明显;0.1 mmol/L的罗格列酮干预24h后,ICAM-1的水平与空白对照组比较,差异无统计学意义(P(0.05);MCP-1荧光强度在罗格列酮干预后也减弱。结论罗格列酮能抑制Hcy诱导的RMECs表达MCP-1和ICAM-1。

Abstract:Objective To investigate the effect of rosiglitazone on the expression of monocyte chemoattractant protein-1 ( MCP-1 ) and intercellular adhesion molecule-1 ( ICAM-1 ) induced by homocysteine(Hcy) in renal microvascular endothelial cells(RMECs). Methods Rat RMECs by primary culture were divided into three groups:blank group, Hcy stimulate group and rosiglitazone intervention group. The expression of MCP-1 ,ICAM- 1 in RMECs was tested by Immunofluorescence and enzyme linked immunosorbent assay respectively. Results The level of ICAM-1 were added significantly( P 〈 0.05 or P 〈 0.01 ) with various concentrations of Hcy, and increased following time and concentrations.The level of ICAM-1 arrived at peak during 24 h to 48 h with 0.1 mmol/L Hcy. The level of ICAM-1 was decreasd significantly, and the higher the concentration of rosiglitazone was,the stronger the in hibitory action was. The level of ICAM-1 in 0.1 mmol/L rosiglitazone group compared with that in blank group was no statistically significance. The fluorescence intensity of MCP-1 was weaken with rosiglitazone. Conclusion Rosiglitazone can inhibit the expression of MCP-1 and ICAM-1 induced by homocysteine in RMECs.
