酸性染料比色法测定鞣苦胶囊中苦参总生物碱的含量

目的建立测定鞣苦胶囊中苦参总生物碱含量的方法.方法采用溶剂萃取酸性染料比色法测定苦参总生物碱含量,以氧化苦参碱为对照品,澳麝香草酚蓝为酸性染料,在pH 7.6缓冲溶液条件下,用氯仿萃取,检测波长417 nm.结果氧化苦参碱在0.698～13.96mg/L范围内与吸光度呈良好线性关系,回归方程为Y=20.243X-1.5136,r=0.9993(n=5),氧化苦参碱的平均回收率为102.67%,RSD=0.74%(n=5).结论所建立方法简便、准确、专属性强.可用于鞣苦胶囊中苦参总生物碱含量测定.

Quantitative Determination of the Total Alkaloids in the Root of Sophora flavescens Used as One of the Ingredients of Rouku Capsules by Acid Dye Colorimetry

Objective To establish a method for quantitative determination of the total alkaloids in the root of Sophora flavescens used as one of the ingredients of Rouku Capsules.Methods The content of the total alkaloids in the root of Sophora flavescens was determined by solvent extraction and acid dye colorimetry, with oxymatrine as a control criterion and bromothymol blue as the acid dye. Under a condition in which the pH value of the buffer solution is equal to 7.6, the total alkaloids in the root of Sophora flavescens were extracted with chloroform, and their content was detected at the wavelength of 417 nm. Results Oxymatrine showed a good linear relationship with absorbance when its concentration was between 0.698 mg/L-13.96 mg/L, and the regression equation established was C=20.243A-1.5136, (r=0.9993, n=5). The recovery of oxymatrine was 102.67% (RSD=0.74%, n=5). Conclusion This procedure, which is easy, accurate and specific, can be used in the quantitative determination of the total alkaloids in the root of Sophora flavescens in Rouku Capsules.
