摘    要:目的了解经典的钙通道阻断剂维拉帕米和肌浆网Ca2 释放抑制剂钌红对去甲肾上腺素(NE)诱导的心肌c-fos、c-myc基因表达的影响.方法通过离体心脏灌流,采用逆转录-多聚酶链反应(RT-PCR)方法,测定左心室c-fos、c-myc表达水平.结果钌红明显降低NE诱导的c-fos、c-myc表达,维拉帕米部分阻断NE诱导的c-fos表达,但对c-myc基因表达无明显影响.结论 NE诱导的心肌c-fos、c-myc基因表达有赖于细胞内钙(Ca2 ];)的增高.

Abstract:Objective To observe the effects of verapamil and ruthenium red on NE induced expression of c fos and c myc in rat ventricle. Methods The mRNA level of c fos and c myc in the left ventricle of the langendorff perfused rat heart were quantitated by reverse transcripition and polymerase chain reaction (RT PCR) under different experiment condition. Results Ruthenium red (10uM)decreased the NE induced c fos mRNA level and c myc mRNA level pronouncedly. Verapamil blacked the effect of NE induced expression of c fos partly, but the c myc mRNA level was unaffected. Conclusion NE induced expression of oncogene c fos and c myc in the ventricle is depended on Ca 2 ] i increase.
