摘    要:目的探讨VCD诱导大鼠卵巢功能衰竭的机制。方法取28天龄雌性SD大鼠,分别用去氧乙烯基环己烯(4.vinylcyclohexenediepoxide,VCD)、芝麻油、生理盐水腹腔注射,观察各组大鼠的动情周期变化;分别于腹腔注射后20天、40天、60天取各组大鼠外周血,用放射免疫法检测FSH、LH、E2、P等激素水平的变化。观察大鼠卵巢及子宫的组织学改变、各级卵泡的数量变化。结果注射VCD60天后,75%的大鼠表现为动情周期紊乱,其卵巢与子宫体积均明显小于对照组;血FSH明显高于对照组(P〈0.05);大鼠卵巢组织中原始卵泡和早期生长卵泡的数量明显减少(P〈0.01)。结论VCD能诱导大鼠卵巢功能衰竭,可作为探讨卵巢早衰发病机制的动物模型。

Abstract:Objective: To assess the incidence of POF, and make an animal model of POF. Methods: Female SD rats (28-days) received identical intraperitoneal injection of three chemicals, VCD, sesame oil and physiological saline, and then the variety of estrous cycle in these three groups was observed. The rats'peripheral blood in these three groups was ob-tained after intraperitoneal injection in 20-days, 40-days and 60 days, to test the diversity of hormone levels (FSH, LH, E2, P). The sections of the rats'ovarian and uterus were made after intraperitoneal injection VCD in 20-days, 40-days and 60-days, and the diversity of histology in rats'ovarian and uterus was then observed, and the number of apoptosis cells and ovarian follicles was counted. Results: In the 60-day after intraperitoneal injection VCD, estrous cycle in disorder was 75% of the rats, and the volume of ovarian and uterus in 60-days group was all atrophied compared with the control group; In the 60-day after intraperitoneal injection VCD, the level of FSH in rats'peripheral blood was significantly higher than that in the control group(P<0.05); The number of primordial follicle and growing follicle in rats'ovarian with VCD injection was sig-nificantly fewer than that in the control group (P<0.01). Conclusion: VCD can induce premature ovarian failure in the rat, which can be an appropriate animal model to study the incidence of POF.
