| 摘 要： | 目的:揭示通幽汤对食管癌抑制作用的细胞内分子机制,确定从功效分类的有效药物及瘀血内结证的分子本质。方法:体外培养食管鳞癌EC9706细胞,通幽汤及活血行气、滋阴养血拆方分别作用于细胞,倒置显微镜观察细胞形态变化;以MTT法检测细胞抑制率,确定最佳药物浓度和作用时间;western blot法观察PI3K/AKT信号通路各蛋白表达变化情况。结果:通幽汤及活血行气拆方可显著抑制EC9706细胞增殖,48h作用最强,通幽汤全方IC50=1523μg.mL-1,活血行气拆方IC50=1001μg.mL-1;可抑制PI3K/AKT信号通路相关蛋白表达,作用强弱依次为:活血行气拆方>通幽汤全方>滋阴养血拆方。结论:通幽汤抑制食管癌细胞增殖主要为活血行气类药物,根据"以方测证"理论,瘀血内结证与PI3K/AKT信号通路表达增强有关。
