青藤碱对小鼠脑缺血再灌注损伤的保护作用

摘    要:目的观察青藤碱腹腔给药对小鼠脑缺血再灌注损伤的保护作用。方法昆明种小鼠随机分成5组,即空白组、模型组、预处理组、青藤碱低剂量组和高剂量组。采用双侧颈总动脉结扎/复灌制备小鼠不完全性全脑缺血再灌注损伤模型;预处理组提前给予夹闭双侧颈总动脉10 s、松开10 s,共重复5次;青藤碱低剂量(40 mg/kg)组、高剂量(80 mg/kg)组腹腔注射给药7 d,观察其对脑缺血再灌注损伤小鼠的脑组织含水量、HE染色后的病理变化和对SOD活性、MDA含量的影响。结果青藤碱高剂量、低剂量组小鼠脑组织含水量均低于模型组;青藤碱减轻小鼠脑细胞的损害程度;青藤碱能降低脑组织MDA含量和提高SOD活性(均P〈0.05)。结论青藤碱腹腔注射给药对小鼠脑缺血再灌注损伤具有保护作用,其作用机制与清除氧自由基有关。

Abstract:Objective To investigate the protective effects and the possible mechanism of Sinomenine on cerebral ischemia-reperfusion injury in mice.Methods Kunming mice were randomly divided into five groups: sham operation group,model group,pretreatment group and two groups pretreated with different doses(40 mg/kg,80 mg/kg).The pretreatment group was made by bilateralligation of common carotid arteries for 10 seconds and reperfusion for 10 seconds,repeating for five times.The Sinomenine pretreatment groups were administered intragastrically with the extract for 7 days,Meanwhile,the other mice were given distilled water.The water content of brain was measured;neural injury of cerebral tissues was observed;malondialdehyde(MDA) levels and superoxide dismutase(SOD) activity glutathione peroxidase(GSH-Px) activity in brain homogenate were detected by using spectrophotometry.Results Compared with I/R group,low and high dose Sinomenine treated group dose-dependently reduced the water content of brain and the damage extent of cerebral cells in mice;sinomenine reduced MDA content in brain tissue and increased SOD activity(all P 0.05).Conclusion Sinomenine has protective effect on cerebral ischemia-reperfusion injury by intraperitoneal injection,and its primary mechanism may be scanvenging oxygen free radicals.
