止血镇痛胶囊对外源性雌激素干预下未成年雌性大鼠卵巢和子宫的影响

目的 探讨止血镇痛胶囊对外源性雌激素干预下未成年雌性大鼠卵巢和子宫的影响。方法 将84只雌性大鼠随机分成空白组和模型组,空白组给予0.5% CMC-Na,模型组给予戊酸雌二醇,灌胃3周后,模型组大鼠随机分成模型对照组、卡巴克络对照组、独一味软胶囊对照组、止血镇痛胶囊低、中、高剂量组。各给药组均灌胃相应药物,空白对照组和模型对照组灌胃生理盐水。7 d后检测大鼠体重、子宫和卵巢脏器指数、血清指标、子宫和卵巢组织病理性以及子宫内膜组织中VEGF表达。结果 与模型对照组比较,止血镇痛胶囊中剂量组大鼠的体重十分明显地增加（P < 0.01）;3组动物的子宫系数均十分明显地减小（P < 0.01）;中、高剂量组大鼠血清中PROG浓度十分明显地升高（P < 0.01）;中、高剂量组大鼠子宫肌层平滑肌细胞透明变性、排列紊乱均减轻（P < 0.05或P < 0.01）;高剂量组大鼠卵巢组织的间质增生明显减轻（P < 0.05）;三组大鼠子宫组织VEGF蛋白表达量均升高（P < 0.05或P < 0.01）。结论 止血镇痛胶囊减轻大鼠子宫肌层平滑肌细胞透明变性、排列紊乱和卵巢组织的间质增生的作用,可能与血清中PROG浓度和子宫组织VEGF蛋白表达量有关,为探讨止血镇痛胶囊的化瘀调经作用提供了依据。

Effects of Zhixue Zhentong Capsules on Ovary and Uterus of Juvenile Female Rats Exposed to Exogenous Estrogen

Objective To investigate the effects of Zhixue Zhengtong Capsules on the ovary and uterus of juvenile female rats under the intervention of exogenous estrogen.Methods A total of 84 female rats were randomly divided into a blank group and a model group. The blank group was given 0.5% CMC-Na, while the model group was given estradiol valerate. After 3 weeks of intragastric administration, the rats in the model group were randomly divided into several groups including a model control group, a Kabak control group, a Duyiwei soft capsule control group, as well as Zhixue Zhengtong Capsules with low, medium and high dose groups. Each drug-administered group was intragastrically administered with the corresponding drugs with the blank control group and the model control group orally administered with normal saline. Seven days later, rat body weight, uterus and ovarian organ index, serum index, uterus and ovarian tissue pathology, as well as VEGF expression in endometrial tissue were measured.Results Compared with the model control group, the weight of the rats in the medium dose group of Zhixue Zhentong Capsules increased significantly (P < 0.01). Uterus coefficients of all the 3 groups were significantly reduced (P < 0.01). The concentrations of PROG in the serum of rats in both middle and high dose groups were significantly increased (P < 0.01). The hyaline degeneration and disorder of the myometrial smooth muscle cells in the medium and high dose groups were alleviated (P < 0.05 or P < 0.01). The interstitial proliferation of ovarian tissue of rats in high dose group was significantly reduced (P < 0.05). The expression of VEGF protein in uterus of all the three groups was increased (P < 0.05 or P < 0.01).Conclusion The effects of Zhixue Zhengtong Capsule on the hyaline degeneration, disorder of arrangement and interstitial proliferation of ovarian tissue in myometrial smooth muscle cells of rats may be related to the concentration of PROG in serum and the expression of VEGF protein in uterus, which offers evidence for further probe into the mechanism of effect of Zhixue Zhengtong Capsules on removal of blood stasis and regulation of menstruation .
