三种血痕DNA抽提方法的比较研究

在用聚合酶链反应技术进行基因扩增研究过程中，采用／氯仿常规方法从人血痕中抽提ＤＮＡ，所抽提ＤＮＡ数量不足用于ＰＣＲ扩增。为提高模板ＤＮＡ的数量，昼避免抽提过程所用试剂对ＰＣＲ的干扰，先后采用裂解法，水煮法抽提ＤＮＡ。结果表明，此两种方法要比酚／氯仿法简便，迅速，效果好。而在后两种方法比较下，水煮法要比裂解法更具优越性，是一种从血痕中提取ＤＮＡ的良好方法。

Comparative Studies on Three Methods of Extracting DNA FROM Bloodstains

During the study of gene amplification ,the quantity of DNA,which was extmcted from the bloodstrain of human In order to increase the quantity of templet DNA,to avoid the distmction of the chemical reagent to PCR during the procedure of chemical extraction, two methods,i.e. lysis and water-boiled,were used.The results show that these two methods are simpler, quicker and more econoli cal than phenol/chlorom methed. The effect Of the two metheds is better than the old one. Comparing the latter two methods,i.e. lysis and water-boiled, water-boiled method is a good way of extracting NDA from bloodstains, it is more advantageous than lysis methed.
