摘    要:目的探讨黄皮酰胺(Clausenamide,Clau)对糖尿病大鼠海马环氧化酶2(cyclooxygenase-2,COX-2)mRNA和蛋白质表达水平的影响。方法♂Wistar大鼠腹腔注射链脲佐菌素(48mg·kg-1)建立糖尿病模型,给予不同药物治疗3mon后,分别以RT-PCR法和免疫组织化学法观察大鼠海马COX-2 mRNA和蛋白质的表达。结果①RT-PCR结果显示,糖尿病大鼠海马的COX-2 mRNA表达明显增加(P<0.01);而Clau(50、25mg·kg-1)治疗组大鼠海马的COX-2mRNA表达明显降低(P<0.01);②免疫组化结果显示,糖尿病大鼠海马COX-2的蛋白质表达明显升高(P<0.01),Clau(50、25mg·kg-1)治疗组大鼠海马的COX-2的蛋白质表达明显降低(P<0.01)。结论黄皮酰胺可能通过抑制海马的COX-2mRNA及蛋白质表达起到糖尿病大鼠海马的保护作用。

Abstract:Aim To explore the effects of Clausena-mide(Clau) on hippocampus cyclooxygenase-2 (COX-2) mRNA and protein expressions in diabetic rats. Methods The diabetic rat model was produced by injecting streptozotocin (STZ,48 mg·kg-1). After 3 months,the COX-2 gene and protein expressions in hippocampus of diabetic rats were detected by RT-PCR and immunohistochemistry respectively. Results ① The results of RT-PCR showed that the expression of COX-2 mRNA in hippocampus of diabetic group rats increased significantly (P<0.01),however,the expression of COX-2 mRNA in the hippocampus of the rats in clau treatment groups (50,25 mg·kg-1) decreased significantly (P<0.01).② The results of immunohistochemistry showed that the protein expression of COX-2 in hippocampus of diabetic group rats increased significantly (P<0.01),but Clau could inhibit the expressions of COX-2(P<0.01).Conclusion Clausenamide can significantly inhibit COX-2 mRNA and protein expressions in hippocampus,which may account for the protective effects.
