活血化瘀药抗大鼠脑缺血-再灌注性血脑屏障损伤的保护作用

摘    要:目的 :探讨活血化瘀药对大鼠局灶脑缺血 再灌注性血脑屏障损伤的保护作用及机制。方法 :采用线栓法制备大鼠大脑中动脉阻塞 (MCAO)模型。SD大鼠 138只 ,♀ ,随机分为对照组、假手术组、灌药 1~ 4组 ,共 6组。缺血 2h ,再灌注 2 4h ,测定大鼠神经功能缺损程度 ;检测血清脑匀浆、血清一氧化氮、C反应蛋白、补体含量及其病理电镜检查。结果 :灌药组大鼠神经功能缺损程度较对照组轻 (P <0 .0 5 ) ;灌药组脑匀浆一氧化氮、血清及脑匀浆C反应蛋白、补体比对照组低 (P <0 .0 5 ) ;灌药组血清 氧化氮较对照组高 (P <0 .0 5 ) ;病理损伤较对照组轻。结论 :活血化瘀药有拮抗大鼠局灶脑缺血 再灌注性血脑屏障损伤的作用。

Abstract:OBJECTIVE To study the protective effect and mechanism of drugs activating blood circulation to remove blood stasis on blood brain barrier injury of ischemic-reinfusion rats.METHODS Animal model of focal cerebral ischemia and reperfusion were made by thread.138 female SD rats were divided into six groups:control group;preudo-surgery group; experimental1,2,3,4, group. The injury degree of neural function and natrate oxide,c-reactive protein;complement.Concentration in serum and homogenate of brain tissue at ischemic 2 h and reinfusion 24 h were determined.The pathologic changes were determined by electron microscope.RESULTS The degree of neural function injury of rats in experimental groups were less than control group ( P < 0.05 ).The concentration of CRP and Complement in serum and homogenate of brain tissue and NO in homogenate of brain tissue in experimental groups were lower than control group.NO concentration in experimental groups were higher than control group ( P < 0.05 ).Pathologic injury in experimental groups were less than control group.CONCLUSIONS Drugs activating blood circulation can decrease the blood brain barrier injury of focal ischemia-reinfusion rats.
