摘    要:目的:观察直流电场对体外培养兔角膜上皮细胞形态及活力的影响。方法:采用组织块培养法培养兔角膜上皮细胞并进行传代培养,将第4代角膜上皮细胞置于强度为8 V/cm的直流电场中暴露4 h,停止电场作用后继续培养12 h;未置于电场中的角膜上皮细胞作为对照。显微成像系统观察细胞形态改变,各时间点培养的细胞行台盼蓝染色计数,比较各组活细胞率。结果:直流电场作用后大部分角膜上皮细胞变长,并垂直于场线方向排列,停止电场暴露后,大多数细胞恢复正常形态及分布。实验组和对照组活细胞率差异无统计学意义(P>0.05)。结论:短时间直流电场作用对培养的兔角膜上皮细胞的形态及活力无明显影响。

Abstract:Objective:To investigate the effects of a direct current electric field(EF) on the morphology and viability of rabbit corneal epithelial cells(rCECs). Methods :The primary rCECs were cultured by tissue block cultivation method and sub-culturing. The P4 sub culture rCECs were exposed to the electric field at 8V/cm for 4 h,and later the cells were continuously cultured for an additional 12 h. Those unexposed cells were used as the control group. The images of the cells were obtained with an image analyzer. The viability of the cells was evaluated by typan blue staining. Results:After exposured to direct current EF for 4 h,many rCECs elongated and oriented to the vector of the field. The cells changed to normal shape and distribution after the exposure ended. The cell viability showed no differ ences between the cells exposed to EF and those in the normal group( P 〉 0.05 ). Conclusion :Short-time exposure to direct current EF do not impair the viability of rCECs.
