上皮性钙黏附蛋白及基质金属蛋白酶-9在皮肤黑色素瘤及色素痣中的表达及意义

摘    要:目的:探讨皮肤黑色素瘤及色素痣中上皮性钙黏附蛋白(E-cadherin)及基质金属蛋白酶-9(matrix metalloproteinase-9,MMP-9)的表达及意义。方法:应用免疫组织化学Envision二步法检测48例皮肤黑色素瘤及20例色素痣组织中E-cadherin与MMP-9的表达,研究其与黑色素瘤进展转移的关系。结果:E-cadherin及MMP-9在皮肤黑色素瘤与色素痣中表达比较,差异均有统计学意义(P<0.01),E-cadherin和MMP-9的表达与肿瘤浸润深度及淋巴结转移有关(P<0.05),二者在黑色素瘤中的表达呈显著负相关(r=-0.45,P<0.01)。结论:MMP-9及E-cadherin表达与黑色素瘤发生、发展有关,MMP-9上调可能与E-cadherin下调共同促进了黑色素瘤的浸润及淋巴结转移。

Abstract:Objective:Investigate the expression and their significance of E-cadherin and MMP-9 in cutaneous melanoma and pigmented nevus. Methods:The expression of E-cadherin and MMP-9 was detected in 48 cases of cutaneous melanoma and 20 cases of pigmented nevus by using envision immunohistochemical technique. Their correlations with metastasis and progression of cutaneous melano- ma were analyzed. Results:The expression differences of the E-cadherin and MMP-9 cutaneous melanoma and pigmented nevus were significant( P 〈 0. 01 ). The expression of E-cadherin and MMP-9 was correlated to the tumour invasion and lymph node metastasis( P 〈 0. 05). There was a negative correlation between E-cadherin and MMP-9 expression in the cutaneous melanoma tissues( r = -0. 45 ,P 〈0. 01). Conclusion: E-cadherin and MMP-9 are important in the tumorigenesis and development of cutaneous melanoma. The increase of MMP-9 and the reduction of E-cadherin are closely related to cutaneous melanoma invasion and lymph node metastasis.
