Rac1、Cdc42在胃癌中的表达与胃癌临床病理特征关系的实验研究

目的探讨Rac1、Cdc42在胃癌组织中的表达和胃癌临床病理特征之间的关系,寻求二者相关性。方法取48例胃癌和12例正常粘膜石蜡标本作S-P免疫组化染色。结果免疫组化证实胃癌组织中Rac1表达显著高于正常胃粘膜(P0.05)。Rac1表达与患者的年龄、性别和肿瘤的生长部位无关;随着病理分期的进展,Ⅲ-Ⅳ期阳性率明显高于Ⅰ-Ⅱ期(P0.05)。低分化组阳性率明显高于中高分化组(P0.05)。胃癌中Cdc42表达显著高于正常胃粘膜(P0.05)。Cdc42表达与患者的年龄、性别和肿瘤的生长部位无关;与肿瘤浸润深度、大小、淋巴结转移有关;随着TNM分期的进展,表达均逐渐增强(各期间P0.05)。结论 Rac1和Cdc42在胃癌组织中表达明显增高,两者在胃癌组织中表达具有显著相关性;二者可能通过多种机制和环节而促进胃癌的发生、进展。

RESEARCH OF EXPRESSION OF Rac1,Cdc42 IN GASTRICC ARCINOMA AND CLINICAL PATHOLOGICAL CHARACTERISTICS OF GASTRIC CANCINOMA

Objective] The expression of Rac1、Cdc42 in gastric cancer were examined to determine whether they are correlated with clinical pathological characteristics of the disease.To investigate the probable mechanism and analyze correlation between Rac1 and Cdc42.Methods] The expression of Rac1 and Cdc42 were detected by SP（Streptavidin-Peroxidase）immunohistochemistry in 12 specimens of normal gastic mucous and 48 specimens of gastric cancer from patients who underwent curative resection.Results] 1.The expression of Rac1 in gastric cancer was more than that in normal gastric tissues（P0.05﹚.Moreover,the expression of Rac1 was correlated significantly with tumor size,depth of local infiltration（P0.05﹚,and not with patient＇s age,sex and the location,distant metastasis of tumor;With regard to the clinical stage,significant differece of Rac1 expression were observed between Ⅲ-Ⅳ stage and Ⅰ-Ⅱ stage（P0.05﹚.2.The expression of Cdc42 in gastric cancer was more than that in normal gastric tissues（P0.05﹚.Moreover,the expression of Cdc42 was correlated significantly with tumor size,depth of local infiltration（P0.05﹚,and not with patient＇s age,sex and the location,distant metastasis of tumor;With regard to the clinical stage,significant differece of Cdc42 expression were observed between Ⅲ-Ⅳ stage and Ⅰ-Ⅱ stage（P0.05﹚.Conclusions]（1）Rac1 and Cdc42 expression in gastric cancer was significntly higher,and correlated with infiltration depth,lymph node metastasis and TNM stage.（2）Moreover,Rac1 expression was related to Cdc42,they maybe have coordinate action and promote each other during gastric cancer occuring,developing,invasion and metastasis.（3）All indicate that Rac1 and Cdc42 are involved in tumorigenesis and progression of gastric cancer they can be regarded as indexes for gastric cancer diagnosis and prognosis evaluation.
