葛根芩连汤及其拆方对UC大鼠结肠平滑肌线粒体跨膜电位的影响

目的 比较葛根芩连汤及其拆方对UC大鼠结肠平滑肌线粒体跨膜电位的影响,探讨葛根芩连汤中哪些药物或药物组合对UC大鼠结肠组织中平滑肌线粒体跨膜电位的影响最明显,并探讨其作用机制.方法 将120只大鼠随机分为8组,采用三硝基本磺酸(TNBS)/乙醇法建立UC模型,分别给予葛根芩连汤及其拆方连续灌胃4W后停药,流式细胞仪测线粒体跨膜电位.结果 模型组大鼠结肠平滑肌线粒体跨膜电位显著低于其它各组.全方组、全方去黄芩组、全方去甘草组明显高于模型组(P＜0.01),其次为去葛根组、全方去黄连组、全方去葛根黄连组(P＜0.05).结论 葛根芩连汤及其拆方中对UC大鼠结肠平滑肌线粒体跨膜电位的影响最明显的药组是葛根、黄连、炙甘草的组合,葛根、黄连、炙甘草的组合具有较好的重建和修复结肠组织黏膜及恢复结肠平滑肌功能的作用.

Effects of Effects of Gegenqinlian Decoction and Their Dissembled Recipes on Smooth Muscle Mitochondrial Transmembrance Potential of Ulcerative colitis Model Rats

objective To compare the effects of Gegenqinlian decoction and their dissembled recipes on smooth muscle mitochondrial transmembrance potential of UC model rats, and to explore which components or the drug combinations in gegenqinlian decoction had the most influence on it, further more to discuss its mechanism. Methods 120 cases were di- vided into eight groups, the ulcerative colitis （UC） rats model were established by trinitrobenzene sulfonic acid or etha- nol. After 4w oral administration with Gegenqinlian decoction and their dissembled recipes by gastrogavage, mitoehondrial transmembrance potential were measured by flow eytometer. Results Smooth muscle mitochondrial transmembrance poten- tial of the rats in model group was significantly lower than that of other groups. Measurement results of mitochondrial trans- membrance potential in the whole decoction group, whole decoction but huangqin group, whole decoction took out gancao group were obviously higher than that of the model group （ P 〈 0.01 ）, followed by removed gegen group, whole decoction but huanglian group, whole decoction took out gegen and huanglian group （ P 〈 0.05）. Conclusions Among the Gegenqinlian decoction and their dissembled recipes, combination group of gegen, huanglian and zhigancao was brought the most obviously influence on the smooth muscle mitochondrial transmembrance potential of UC model rats. It proved that the combination group has significant effect on reconstruction and restoration of the colon tissue mucosa and recovery the fuc- tion of colon smooth muscle.
