新康泰营养包对心肌线粒体钙和细胞色素C及Caspase-3活性的影响

摘    要:目的研究新康泰营养包对过度训练大鼠心肌线粒体钙浓度和细胞色素C及Caspase-3活性的影响,以探讨新康泰营养包抗过度训练大鼠心肌细胞凋亡影响的作用机理。方法以过度训练大鼠为运动疲劳模型,用Daly方法、微电极法测定细胞色素C,用德国Boehringer Mannheim公司提供的试剂盒测定心肌细胞中Caspase-3的活性。结果过度训练可造成心肌线粒体钙超载,而新康泰营养包具有抑制心肌钙超载的作用;过度训练可导致大鼠心肌细胞中Caspase-3活性显著升高,而新康泰营养包可抑制过度训练大鼠心肌细胞中Caspase-3活性显著升高。过度训练组和过度训练+蒸馏水组大鼠心肌组织中细胞色素C含量明显高于对照组和过度训练+新康泰营养包组大鼠(P〈0.01)。结论新康泰营养包具有防止过度训练造成的心肌细胞凋亡增加,防止过度训练造成的心肌组织中钙超载,从而有效地阻止心肌线粒体内钙超载,保护心肌线粒体膜的功能,使得心肌线粒体内细胞色素C不漏入胞浆,从而使心肌组织中Caspase级联反应不被启动,有效地阻止了心肌细胞凋亡发生。

Abstract:Objective To explore the mechanism of the XINKANGTAI eliminating exercise-induced apoptosis of cadiocyte in rats by observing the density of calcium and cytochrome-C and the activity of Caspase-3 in the mitochondrion of myocardium of rats.Methods Daly methods and microelectrode method were used to determine the density of calcium and cytochrome-C,and kit made by Boehringer Mannheim of Germany for the determination of the activity of Caspase-3 in rat model of exercise-induced fatigue.Results Compared with the training rats in control group,the calcium overload of the avtivity of Caspase-3 and the advance of cytochrome C in cardiac muscle were inhibit significantly with the use of XINKANGTAI in the training group (P<0.01).Conclusion XINKANGTAI can prevent the cascade activation of Caspase in cardiac muscle by restraining the calcium overload,reducing the apoptosis of cardiac muscle induced by overtraining.
