卡介苗、卡介苗多糖核酸对肺炎大鼠肺Toll样受体mRNA的影响

摘    要:目的探讨卡介苗多糖核酸(BCG-PSN)、卡介苗(BCG)对肺炎大鼠肺TLR2、TLR4mRNA表达的影响。方法57只大鼠随机分为肺炎模型组(PM组,n=51)及健康对照组(HC组,n=6).第4天PM组随机抽取6只(非吸入组,NI组)进行相关检查确定肺炎模型是否制作成功。然后肺炎模型组45只肺炎大鼠再随机分成生理盐水组(NS组)、卡介苗组(BCG组)、卡介苗多糖核酸组(BCG-PSN组)。每组又按吸入次数分为3次、5次、7次组。采用RT-PCR方法研究肺炎大鼠肺TLR2mRNA、TLR4mRNA的表达,以及肺炎时雾化吸入前后肺TLR2mRNA、TLR4mRNA表达。结果肺炎第4天,肺TLR2mRNA、TLR4mRNA表达与HC组相比无统计学差异。吸入BCG-PSN5次时,TLR2mRNA表达与HC组相比P〈0.05,7次时P〈0.01;与NI组及NS组相比吸入BCG-PSN7次时P〈0.05。吸入BCG3次时,TLR2mRNA表达与HC组、NI组、NS组相比,P〈0.05,吸入5次、7次时,P〉0.05。同组间吸入BCG或BCG-PSN5次,7次时TLR4mRNA表达略增强,但无统计学差异(P〉0.05)。结论BCG、BCG-PSN能使肺炎大鼠肺TLR2mRNA表达明显增强,但BCA3-PSN较BCG起效慢、作用强、持续时间长,而且也不能使肺炎大鼠肺TLR4mRNA表达增强。
